Structural characterisation of Peptide-bound SARS-CoV-2 Nsp9 RNA-replicase 3C sequence forms sheet interactions with the N terminus of the protein from the other protomer, as a result N terminal residues move outward by 1.6 (residues 610). The significance of this is yet to be established.

Virtual screening of 2985 FDA approved drugs using in silico methods revealed 5 potential compounds (Alvesco, troglitazone, avodart, dihydroergotoxine, and fluspirilene) against SARS-CoV-2 NSP9. The PCA analysis result suggested that avodart formed a slightly more stable complex compared to the other complexes.

Nsp13-1 stabilizes SARS-CoV-2 replication and transcription (RTC) complex by contacting with nsp13-2, which anchors the 5'-extension of RNA template, and interacting with nsp7-nsp8-nsp12-RNA. Different orientations of nsp13-1 results in different interactions with the two forms of mini RTC. Mini RTC with an nsp12R365A mutation has greater helicase activity compared to individual apsnsp13. Nsp13-1T216A mutation has a decreased helicase activity.
